Bovine landscape genetics in Brazil Scientia Agricola
Souza,Ana Clara Barbosa de; Egito,Andréa Alves do; Peripolli,Vanessa; McManus,Concepta Margaret.
ABSTRACT The aim of this study was to explore spatial genetic structure patterns in cattle breeds adapted to local conditions in Brazil. We georeferenced 876 animals of ten breeds raised in Brazil kept in the Genebank of Brazilian Agricultural Research Corporation (Embrapa) by sample locations using the QGIS 2.4.0 software. The Mantel tests, spatial autocorrelation, and Monmonier tests were performed. The distances for spatial correlation tests ranged from 5 to 15 classes. The results indicated genetic discontinuities in cattle breeds from the Midwest, South, and Southeast of the country. Correlation between genetic and geographic distance was low, but significant. The Monmonier Maximum Distance Algorithm indicated an initial subdivision of Curraleiro and...

D-loop haplotype diversity in Brazilian horse breeds Genet. Mol. Biol.
Ianella,Patrícia; Albuquerque,Maria do Socorro Maués; Paiva,Samuel Rezende; Egito,Andréa Alves do; Almeida,Leonardo Daniel; Sereno,Fabiana T. P. S.; Carvalho,Luiz Felipe Ramos; Mariante,Arthur da Silva; McManus,Concepta Margaret.
Abstract The first horses were brought to Brazil by the colonizers after 1534. Over the centuries, these animals evolved and adapted to local environmental conditions usually unsuitable for exotic breeds, thereby originating locally adapted Brazilian breeds. The present work represents the first description of maternal genetic diversity in these horse breeds based on D-loop sequences. A D-Loop HSV-I fragment of 252 bp, from 141 horses belonging to ten Brazilian breeds / genetic groups (locally adapted and specialized breeds) were analysed. Thirty-five different haplotypes belonging to 18 haplogroups were identified with 33 polymorphic sites. Haplotype diversity (varying from 0.20 to 0.96) and nucleotide diversity (varying from 0.0039 to 0.0239) was lower...

Genetic association between longevity and linear type traits of Holstein cows Scientia Agricola
Kern,Elisandra Lurdes; Cobuci,Jaime Araujo; Costa,Cláudio Napolis; McManus,Concepta Margaret; Braccini Neto,José.
Longevity is a desirable trait in the dairy industry because of its relationship to profitability. The aim of this study was to estimate genetic parameters for longevity measurements related to productive life, or life in the herd, and linear type traits of Brazilian Holstein cows born between the years 1990 and 2008. The (co) variance components were estimated by the restricted maximum likelihood method. The heritability for measurements of longevity and linear type traits ranged from 0.05 to 0.07 and 0.08 to 0.39, respectively. The genetic correlations between measurements of longevity and linear type traits ranged from -0.39 to 0.31. Direct selection for longevity does not necessarily lead to long-lived cows, due to low heritability. Indirect genetic...

Genetic characterization of coat color genes in Brazilian Crioula sheep from a conservation nucleus PAB
Cavalcanti,Lilian Cristina Gomes; Moraes,José Carlos Ferrugem; Faria,Danielle Assis de; McManus,Concepta Margaret; Nepomuceno,Alcebiades Renato; Souza,Carlos José Hoff de; Caetano,Alexandre Rodrigues; Paiva,Samuel Rezende.
Abstract: The objective of this work was to identify single nucleotide polymorphisms (SNPs) in resequencing data from MC1R, ASIP, and TYRP1 genes derived from Crioula sheep (Ovis aris) with different coat colors. Polymorphisms in the ASIP (agouti-signaling protein), MC1R (melanocortin 1 receptor), and TRYP1 (tyrosinase-related protein 1) genes were analyzed in 115 sheep from Embrapa’s conservation nucleus of crioula sheep, in Brazil. A total of 7,914 bp were sequenced per animal, and 14 SNPs were identified. Two additional assays were performed to detect duplications and deletions in the ASIP gene. Ninety-five percent of the coat color variation was explained by epistatic interactions observed between specific alleles in the MC1R and ASIP genes. Evidence...

Genetic parameters for five traits in Africanized honeybees using Bayesian inference Genet. Mol. Biol.
Padilha,Alessandro Haiduck; Sattler,Aroni; Cobuci,Jaime Araújo; McManus,Concepta Margaret.
Heritability and genetic correlations for honey (HP) and propolis production (PP), hygienic behavior (HB), syrupcollection rate (SCR) and percentage of mites on adult bees (PMAB) of a population of Africanized honeybees were estimated. Data from 110 queen bees over three generations were evaluated. Single and multi-trait models were analyzed by Bayesian Inference using MTGSAM. The localization of the hive was significant for SCR and HB and highly significant for PP. Season-year was highly significant only for SCR. The number of frames with bees was significant for HP and PP, including SCR. The heritability estimates were 0.16 for HP, 0.23 for SCR, 0.52 for HB, 0.66 for PP, and 0.13 for PMAB. The genetic correlations were positive among productive traits...

Isolation and characterization of mesenchymal stem cells derived from bovine Wharton's jelly and their potential for use in cloning by nuclear transfer Ciência Rural
Silva,Carolina Gonzales da; Martins,Carlos Frederico; Cardoso,Tereza Cristina; Cunha,Elisa Ribeiro da; Bessler,Heidi Christina; McManus,Concepta Margaret; Pivato,Ivo; Báo,Sônia Nair.
ABSTRACT: Wharton's jelly is a source of mesenchymal stem cells (MSCs) that had not yet been tested for bovine embryo production by nuclear transfer (NT). Thus, the objective of this study was to isolate, characterize and test MSCs derived from Wharton's jelly for embryo and pregnancy production by NT in cattle. The umbilical cord was collected during calving and cells derived from Wharton's jelly (WJCs) were isolated by explant and cultured in Dulbecco's Modified Eagle Medium. Skin Fibroblasts (FB) were isolated after 6 months of life. Morphological analysis was performed by bright field and scanning electron microscopy (SEM) during cell culture. Phenotypic and genotypic characterization by flow cytometry, immunocytochemistry, RT-PCR and differentiation...

The objective of the present study consisted of describing dam and calf suckling behaviour of Curraleiro Pé Duro cattle. In this study, 38 mother-offspring pairs and one mother-offspring-orphan trio were observed for 10 hours daily during three consecutive days spaced every four months over a period of one year. After identification, the animals were observed under field conditions where calf posture and the number of suckling episodes (NS), mean suckling duration (MSD), total suckling duration per day (TSD) as well as natural weaning of these animals were recorded. The model assessed the effects of calf sex and age as well as feeding time. Suckling episodes (70.6%) had a duration of one to five minutes and the calf that suckled in the inverted parallel...

Modelling beef cattle production systems from the Pampas in Brazil to assess intensification options Scientia Agricola
Lampert,Vinícius do Nascimento; Canozzi,Maria Eugênia Andrighetto; McManus,Concepta Margaret; Dill,Matheus Dhein; Oliveira,Tamara Esteves de; Mercio,Thomaz Zara; Teixeira,Odilene de Souza; Barcellos,Júlio Otávio Jardim.
ABSTRACT Traditional livestock productivity is hard to estimate, since it depends on a gamut of animal production indicators that are difficult to measure for many farms. Thus, we propose an analytical method for estimating productivity and understanding the importance of animal production indicators under different full-cycle cattle production systems in Brazil. To evaluate the impact of these indicators, equations were derived from a comutational model of herd evolution for estimating the output parameters of the system as follows: productivity per hectare (PH) and offtake rate (COR), as a function of the indicators; calving rate (CR), mating age (AM), age of slaughter (AS) and stocking rate (SR). For this analysis, twenty-seven scenarios (simulation...

Validation of a microsatellite panel for parentage testing of locally adapted and commercial goats in Brazil Genet. Mol. Biol.
Silva,Elizabete Cristina da; McManus,Concepta Margaret; Guimarães,Maria Pia Souza Lima Mattos de Paiva; Gouveia,Aurora M.G.; Facó,Olivardo; Pimentel,Daniel M.; Caetano,Alexandre Rodrigues; Paiva,Samuel Rezende.
Brazilian goats are generally kept in small herds and extensive rearing systems, mainly in the northeastern region of the country. Despite production improvement in recent years, the lack of pedigree control has affected genetic progress. This study aimed to validate a panel of 16 microsatellites for parentage testing in locally adapted and commercial goats breeds raised in Brazil, as well as to compare its efficiency with the panel recommended by the Brazilian Ministry of Agriculture, Livestock and Supplies (MAPA) in 2004. The number of alleles and expected heterozygosity (He) per marker ranged from four to 18, and from 0.051 to 0.831, respectively. Using all markers, 100% of parentage cases of the validation dataset were resolved with a strict confidence...
